Multipotentiality of Single Embryonal Carcinoma Cells.

چکیده
In order to test the hypothesis that embryonal carcinoma cells are multipotential stem cells of a teratocarcinoma, an in vivo cloning technic was designed. Small embryoid bodies containing mostly embryonal carcinoma were obtained from ascitic con version of a murine teratocarcinoma and were dissociated with trypsin to form a sus pension of single cells; the single cells were picked up in small capillary tubes and transplanted directly into mice. From over 1700 single cell grafts, 44 clonal lines were obtained; 43 of these lines were teratocarcinomas composed of as many as fourteen well differentiated somatic tissues in addition to embryonal carcinoma. These 43 lines varied in their degree of differ entiation, capacity to produce embryoid bodies, and in growth rate. The remaining clonal line showed limited potential for differentiation, producing only yolk sac, trophoblast, and embryonal carcinoma. The results demonstrated the multipotentiality of single embryonal carcinoma cells, as well as the heterogeneity of the embryonal carcinoma of a teratocarcinoma. The capacity of single embryonal carcinoma cells to differentiate into benign tissues sup ports neither the dogma of the irreversibility of the malignant transformation nor the somatic cell mutation theory of cancer. These findings were interpreted as giving strong support to the stem cell theory of cancer.

Dehydroepiandroesteron increased proliferation of neural progenitor cells derived from p19 embryonal carcinoma stem cells.
Introduction: The p19 line of embryonal carcinoma cells develops into neurons, astroglia and fibroblasts after aggregation and exposure to retinoic acid (RA). Dehydroepiandroesteron (DHEA) is a neurosteroid, can increase proliferation of human neural stem cell (NSC) and positively regulated the number of neurons produced. This study was initiated to assess the effect of DHEA on neural progenito...
متن کاملThe Ultrastructure of Primordial Germinal Cells of the Fetal Testes and of Embryonal Carcinoma Cells of Mice.
This study compares the ultrastructure of murine embryonal carcinoma cells, the multipotential stem cells of teratocarcinomas, with the constituents of the primordial testicular tubules in which they arise. The primordial tubules were composed of Sertoli cells and primordial germinal cells ; the former were well differentiated with Golgi membranes and well developed cisternal endoplasmic reticu...
متن کاملP-88: Comparing Epigenetic Profile of Oct4 Regulatory Region in Embryonal Carcinoma Cells under Retinoic Acid Induction
Background: Embryonal carcinoma (EC) cells derived from germ cell tumors are valuable tools for investigating differentiation and developmental biology processes in vitro. The advantage of the reproducible and rapid expansion of these cell lines provides a useful alternative to embryos for the study of mammalian cell differentiation. During early stages of cell differentiation, the rate of tran...
متن کاملمهار بیان ژن GFP به وسیله تداخل RNA (RNAi) در دودمان سلولی کارسینومای جنینی P19
Introduction: RNA interference (RNAi) is a phenomenon of gene silencing that uses double-stranded RNA (dsRNA), specifically inhibits gene expression by degrading mRNA efficiently. The mediators of degradation are 21- to 23-nt small interfering RNAs (siRNA). The use of siRNAs as inhibitors of gene expression has been shown to be an effective way of studying gene function in mammalian cells. Ai...
متن کاملImmature Teratoma with Embryonal Carcinoma; a Rare Malignant Mixed Germ Cell Tumor in a 13-Year-Old Girl
Malignant mixed germ cell tumors, though rare overall, are the most common type of malignant ovarian neoplasms in young and adolescent girls. These tumors are rapidly growing and can metastasize. We report a case of 13-yr-old girl who presented at SHKM GMC, Nalhar, Mewat, Haryana, India in December 2013 with huge abdominal lump of a malignant mixed germ cell tumor comprising both immature terato...
